Laurate can be produced in the seed reserve oil of Brassica napus (rapeseed) by the expression of an heterologous lauroyl acyl-carrier protein thioesterase under the control of a napin seed-storage protein promoter. Analysis of a large number of transgenic events, and their progeny after self-pollination, shows that laurate can accumulate to nearly 60% of the triglyceride acyl groups. Up to 40 mol% laurate the phenotype is correlated positively with the number of thioesterase gene copies. The use of a tandem gene construct elevates the average laurate content. This effect correlates with an increased average number of T-DNA insertions per event; no cis -inactivation of tandem genes is apparent. Above 40 mol% laurate other factors apparently limit the phenotype. The expression timing conferred by the napin promoter is unlikely to be limiting, as it covers almost the entire period of oil deposition. A more significant limitation resides in the second acylation reaction of oil biosynthesis, as shown by the very low incorporation of laurate at the sn -2 acyl group. The novel, high-laurate oil is consequently rich in sn -1,3-dilauroyl triglycerides, but its unusual composition appears to pose no problems for mobilization during seed germination. 相似文献

The Lower Cambrian Chengjiang fauna is reviewed and shown to be closely comparable with the younger Burgess Shale fauna. but with various differences in detail. A diverse group of more or less annulated lobopod animals including 'armoured lobopods' are regarded as representatives of the phylum Onychophora. 'Trilobitomorphs' include several new types. Probable protaspides of the trilobitomorph Naraoia are described. No molluses or deuterostomes have been identified. The preservational orientations of the various taxa are reviewed and compared with orientations of the Burgess Shale taxa. Orientation in the sediment is found to be closely correlated to the original shape of individuals. Several new genera and species are described: the segmented. worm-shaped Yunnanozoon lividum gen. et sp.n., the 'armoured lobopods' Onychodictyon ferox gen. et sp.n. and Cardiodictyon catenulum gen. et sp.n. and the arthropods Saperion glumaceum gen. et sp.n., Sinoburius Iunaris gen. et sp.n., and Xandarella spectaculum gen. et sp.n. 相似文献

While much work in political science has examined the impact of racial cues on individual perceptions, we know little about how individuals evaluate members of minority outgroups on issues that are not linked to stereotypes. We measure the impacts of Hispanic and White cues on individual assessments related to a stereotype-independent norm violation: alcoholism. We test three competing theories – cognition, intergroup emotions, and social identity – using a population-based vignette experiment included in the General Social Survey. Our results contradict much of the literature, but keep with social identity theory''s predictions. Hispanic alcoholics, when Hispanics constitute the outgroup, are assessed less negatively than White alcoholics in the ingroup, the latter experiencing what is called the black sheep effect. The black sheep effect occurs when ingroup members are more punitive towards members of the ingroup than the outgroup. However, the black sheep effect does not extend to measures that are more consistent with outgroup stereotypes, such as violence or money mismanagement; Hispanic alcoholics are evaluated more negatively than Whites on these measures. The implication is that the effect of racial cues depends strongly on issue linkages to group stereotypes. 相似文献

The last few years have seen the identification of numerous small molecules that selectively inhibit specific class I isoforms of PI3K (phosphoinositide 3-kinase), yet little has been revealed about the molecular basis for the observed selectivities. Using site-directed mutagenesis, we have investigated one of the areas postulated as being critical to the observed selectivity. The residues Thr(886) and Lys(890) of the PI3Kgamma isoform project towards the ATP-binding pocket at the entrance to the catalytic site, but are not conserved. We have made reciprocal mutations between those residues in the beta isoform (Glu(858) and Asp(862)) and those in the alpha isoform (His(855) and Gln(859)) and evaluated the potency of a range of reported PI3K inhibitors. The results show that the potencies of beta-selective inhibitors TGX221 and TGX286 are unaffected by this change. In contrast, close analogues of these compounds, particularly the alpha-isoform-selective compound (III), are markedly influenced by the point mutations. The collected data suggests two distinct binding poses for these inhibitor classes, one of which is associated with potent PI3Kbeta activity and is not associated with the mutated residues, and a second that, in accord with earlier hypotheses, does involve this pair of non-conserved amino acids at the catalytic site entrance and contributes to the alpha-isoform-selectivity of the compounds studied. 相似文献

The KwaZulu-Natal region of South Africa hosts a large diversity of asclepiads (Apocynaceae: Asclepiadoideae), many of which are endemic to the area. The asclepiads are of particular interest because of their characteristically highly evolved floral morphology. During 3 months of fieldwork (November 2000 to January 2001) the flower visitors and pollinators to an assemblage of nine asclepiads at an upland grassland site were studied. These observations were augmented by laboratory studies of flower morphology (including scanning electron microscopy) and flower colour (using a spectrometer). Two of the specialized pollination systems that were documented are new to the asclepiads: fruit chafer pollination and pompilid wasp pollination. The latter is almost unique in the angiosperms. Taxa possessing these specific pollination systems cluster together in multidimensional phenotype space, suggesting that there has been convergent evolution in response to similar selection to attract identical pollinators. Pollination niche breadth varied from the very specialized species, with only one pollinator, to the more generalized, with up to ten pollinators. Pollinator sharing by the specialized taxa does not appear to have resulted in niche differentiation in terms of the temporal or spatial dimensions, or with regards to placement of pollinaria. Nestedness analysis of the data set showed that there was predictability and structure to the pattern of plant-pollinator interactions, with generalist insects visiting specialized plants and vice versa. The research has shown that there is still much to be learned about plant-pollinator interactions in areas of high plant diversity such as South Africa. 相似文献

New material of early growth stages of the Silurian (Llandovery) trilobite Acernaspis is described. Pre-adult ontogenetic stages of this genus closely resemble adults of the post-Llandovery genus ***Ananaspis. A heterochronic descent of Ananaspis from Acernaspis is proposed. ***Ananaspis is interpreted as pae-domorphic, having arisen largely through neoteny. A range of short-chain aliphatic monocarboxylates, both unsubstituted and substituted with hydroxy, chloro and keto groups, were shown to inhibit transport of L-lactate and pyruvate into both guinea-pig cardiac myocytes and rat erythrocytes. The carrier of heart cells exhibited a higher affinity (approx. 10-fold) for most of the monocarboxylates than did the erythrocyte carrier. A notable exception was L-lactate, whose Km for both carriers was similar. The K1 values of the two carriers for inhibitors such as phenylpyruvate and alpha-cyanocinnamate derivatives were also different. The high affinity of the heart cell carrier for ketone bodies and acetate may be physiologically important, since these substrates are used as fuels by the heart. 相似文献

A procedure has been described which is applicable to the screening of large numbers of samples for anticholinesterase activity. The method is sufficiently sensitive to detect cholinesterase depression due to as little as 30 ppt paraoxon; 3 ppb malaoxon or 2 ppm of carbaryl in either ethylene glycol or water. While the method does not allow the absolute differentiation between compounds which might be present, some classification is possible, i.e., for compounds which must be activated before they possess anticholinesterase activity. 相似文献
